Serial dilution testing is a traditional culture-based method used to estimate viable microbial populations such as sulfate-reducing bacteria (SRB), acid-producing bacteria (APB), iron-reducing bacteria (IRB), and general heterotrophic bacteria (GHB). Samples are inoculated into anaerobic media and incubated to observe microbial growth over a testing period of days/weeks/months.
OSP media is produced in accordance with NACE Standard TM0194-2014 for serial dilution testing and can be manufactured to meet API-38 specifications at various salinities. Media is prepared under strict anaerobic conditions without sulfite-based oxygen scavengers to maintain quality and consistency.
Culture-based methods detect only viable organisms capable of growth under the provided laboratory conditions. They require incubation time, may underestimate total populations, and can miss microbes that are active but not easily cultured. For this reason, many operators supplement serial dilution testing with ATP or genetic methods for a more comprehensive microbial assessment.
Each testing method provides different but complementary information. ATP testing offers rapid insight into the total active microbial load. qPCR provides targeted quantification of specific functional groups associated with souring and corrosion. DNA 16S sequencing delivers broad community characterization and relative percentages of each. Serial dilution testing satisfies traditional monitoring requirements and regulatory standards. When used together, these tools create a comprehensive microbial monitoring strategy that supports informed decision-making and long-term asset protection.
